Biosynthesis of Nondigestible Galactose-Containing Hetero-oligosaccharides by <i>Lactobacillus plantarum</i> WCFS1 MelA ?-Galactosidase

This work describes the high capacity of MelA ?-galactosidase from Lactobacillus plantarum WCFS1 to transfer galactosyl residues melibiose C6-hydroxyl group disaccharide-acceptors with ?-linkages (lactulose, lactose, and cellobiose) or ?-linkages (isomaltulose isomaltose) produce novel galactose-containing hetero-oligosaccharides (HOS). A comprehensive nuclear magnetic resonance characterization products derived melibiose:lactulose reaction mixtures revealed biosynthesis ?-d-galactopyranosyl-(1 ? 6)-?-d-galactopyranosyl-(1 4)-?-d-fructose as main component well presence 3)-?-d-galactopyranosyl-(1 6)-?-d-galactopyranosyl-(1 4)-?-d-fructose. Melibiose-derived ?-galactooligosaccharides (?-GOS), manninotriose verbascotetraose, were also simultaneously synthesized. An in vitro assessment intestinal digestibility biosynthesized HOS a resistance ?-galactosides lactulose, cellobiose, isomaltulose. According evidence gathered for conventional ?-GOS certain disaccharides used acceptors this work, these nondigestible could be potential candidates selectively modulate gut microbiota composition, among other applications, such low-calorie food ingredients.
